❗ Despite advances in physiologically based modeling, many approaches still assume homogeneous mixing of drug and gastric contents. Yet, real clinical data tell a different story—irregular plasma profiles, multiple secondary peaks, prolonged lag times, or unexpectedly early Cmax values.
🤔 These phenomena point to a much more complex reality: non-uniform drug distribution in the stomach and the critical role of the Magenstrasse (stomach road), especially in the presence of gastric secretions and repeated water intake.
